Cyclin-dependent kinase 7 (CDK7) is a member of CDK family which is involved in cell cycle progression and transcriptional regulation. Therefore, deregulation of CDK7 has been observed in various types of cancer including ovarian cancer and advanced solid tumors. This potentiates CDK7 as a valid therapeutic target for the treatment of cancer. Thus, selective inhibition of CDK7 can lead to the development of new anticancer therapy with less side effects. However, it is very difficult to selectively inhibit CDK7 as the structure of CDK7 is similar with other CDKs along with the conserved ATP binding site. Despite that, several CDK7 selective inhibitors like SY-5609, Q901, XL102, LGR6768 have reached clinical phases. In this review work, we have compiled updated information on the biology and function of CDK7, its involvement in cancer. Furthermore, we have also studied existing inhibitors of CDK7 which are in various phases of preclinical and clinical trials. Rubina Haque Sorno B.
