Cataloged from the PDF version of the thesis. Includes bibliographical references (pages 50-59). SIRT6 is one of the sirtuin family members, a kind of NAD+-dependent histone deacetylase and ADP-ribose transferase enzyme. It controls aging, cancer, obesity, insulin resistance, inflammation, and energy metabolism, playing a significant role in both physiological and pathological processes. Sirt6 levels and function diminish with age and overnutrition, two key risk factors for obesity and diabetes, which results in improper glucose and lipid metabolism. Mice with SIRT6 ablation across the body have acute hypoglycemia. SIRT6 deficiency enhances diet-induced obesity and insulin resistance and causes liver steatosis. Diabetes and obesity are protected against SIRT6.This review examines the data supporting SIRT6's emerging function as a regulator of metabolism in mammals and lists its key roles in obesity and diabetes.
