Catalogued from PDF version of thesis. Includes bibliographical references (pages 49-53). The experiment started with collecting samples of Pseudomonas aeruginosa from a clinical source of burnt patients and storing them in T1N1 media at room temperature. For confirmation of identification of the supplied organism Pseudomonas aeruginosa, they were grown in different selective media, and related biochemical tests were carried out for its identification. Next, the ability of the organism to form biofilm was determined. Qualitative and quantitative methods were carried out to see if our tested organism could form biofilms. After successfully forming biofilms with the tested organism, we applied ascorbic acid solutions in different concentrations to assess the effectiveness of vitamin C in reducing the ability of these microorganisms to inhibit biofilm formation. Antibiotic susceptibility tests were also performed for the tested organism and out of six isolates used, all of them were resistant to at least seven antibiotics of three different classes. As Pseudomonas aeruginosa is resistant to antibiotics and there are concerns about its inefficacy of antibacterial medications, alternative ways can be considered to combat such microorganisms. In this study, it was observed that some concentration of ascorbic acid was able to reduce the ability of biofilm formation by Pseudomonas aeruginosa. From this study, it was found that Vitamin C at 25.6 μg/mL of concentration in the microtitre plate method and at 50 μg/ /mL concentration in the tube method could inhibit biofilm formation.
